Two parts of one organism

A fungus spends most of its life as mycelium: a web of fine threads that spreads through soil, wood or, in a farm, a bag of sterilised grain. When conditions are right it produces a fruiting body, the cap-and-stem structure we call a mushroom. Both parts are the same organism, but they are not the same ingredient once they are dried and powdered.

Why "mycelium on grain" can be mostly grain

Fruiting bodies are harvested, dried and ground, so the powder is mushroom. Mycelium is different. The commercial method is to inoculate cooked grain — rice, oats, sorghum — and let the threads colonise it. Separating mycelium from grain is impractical, so the whole block is dried and milled together. The result, sometimes labelled "mycelial biomass" or "myceliated grain," contains mycelium plus whatever grain was not consumed, which can be most of the weight.

That matters for dosing. If a label promises 500 mg of "lion's mane" and the ingredient is myceliated rice, an unknown share of that 500 mg is rice starch. The label is technically accurate and practically misleading.

Mycelium isn't worthless

It would be easy to conclude that fruiting body is always better, and that is the marketing line of several brands. The evidence is more nuanced. Memorial Sloan Kettering's lion's mane summary lists a pilot trial in early Alzheimer's disease that used mycelium enriched for erinacine A, a compound found mainly in mycelium rather than the fruiting body. Mycelium grown in liquid culture, without grain, can be a legitimate ingredient. The problem is specifically undisclosed grain, not mycelium as such.

Beta-glucan is the comparable number

Beta-glucans are the cell-wall polysaccharides that most of the immune research on mushrooms measures. Crucially, they can be counted. McCleary and Draga published a validated enzymatic method in the Journal of AOAC International that measures total glucan and alpha-glucan separately, so the beta-glucan fraction is the difference. Alpha-glucan is essentially starch and glycogen — the signature of grain. A powder that tests high in beta-glucan and low in alpha-glucan is mushroom tissue; one that tests the reverse is mostly its substrate.

This is why we rate label transparency in our methodology partly on whether a brand publishes beta-glucan. It is the one number that lets you compare a fruiting-body extract against a mycelium product on equal terms.

What does beta-glucan percentage tell you?

Beta-glucans are the polysaccharides most mushroom immune research measures, and they can be quantified with a validated enzymatic assay that separates them from alpha-glucans like starch. A high beta-glucan and low alpha-glucan result indicates real mushroom tissue; the reverse indicates grain. A brand that publishes beta-glucan per serving lets you compare across products.
